## Challenge

Although current drug discovery has progressed significantly, drug delivery continues to remain a major hurdle, with many promising solutions failing because they cannot reach their intended targets safely or efficiently. Current delivery technologies, including LNPs, AAVs, and ADCs, face fundamental limitations such as toxicity, redosing challenges, and the extrahepatic barrier, which makes targeting tissues beyond the liver difficult. At the same time, while cell-derived nanoparticles offer strong biological potential, they have traditionally remained difficult to control, scale, and manufacture consistently, creating a major barrier to their industrialization.

SHIFTBIO is addressing this gap by **transforming** these biological particles into a **scalable, pharma-grade precision delivery** infrastructure designed to unlock new treatments for diseases previously deemed incurable.

## Disruptive Innovation

SHIFTBIO’s disruptive innovation is a programmable Natural Nanoparticle (NNP) platform that transforms cell-derived nanoparticles into scalable, pharma-grade drug delivery systems. Its proprietary NSM scaffold enables nanoparticles to be engineered with specific targeting motifs and therapeutic cargoes, while AI-driven modeling helps predict and optimize their design before testing. Combined with a scalable manufacturing process, this approach aims to enable more precise delivery of complex therapies to previously difficult-to-target tissues.

## Impact

SHIFTBIO’s precision drug delivery platform could transform the development of treatments for diseases with limited or no therapeutic options. The company aims to improve access to medicines by enabling safer, more precise, and efficient delivery of complex therapies, particularly for challenging diseases affecting the brain and immune system.

## Leveraging the 3DEXPERIENCE Platform

SHIFTBIO plans to use the 3DEXPERIENCE platform as a catalyst to accelerate their development journey and move even faster than a traditional "smart team". The company intends to leverage the platform's capabilities in several key areas:

- **Simulation and Precision**: To model cell-derived nanoparticles before they are physically built to ensure that the resulting drug delivery is more powerful and precise.
- **Manufacturing Optimization**: They plan to further improve the manufacturing processes for their new therapeutic modalities in the near future by utilizing specific tools within the platform, namely BIOVIA and DELMIA.
- **Scientific Insight and AI Training**: Utilizing BIOVIA & previous research to identify essential key residues and interfaces between their NSM scaffold and specific proteins

## Collaborative & Collective Intelligence

SHIFTBIO leverages collective intelligence through a multidisciplinary team combining expertise in nanoparticle biology, protein engineering, AI-based design, and manufacturing. The company also works closely with research partners, including VIB and the BIOVIA content research team, to understand the interactions between its NSM scaffold and specific proteins. This collaborative research has helped build the mechanistic understanding behind its nanoparticle engineering and predictive AI models.
